All detailed in the last story arc of The Clone Wars. Midichlorians are Living Force batteries, receiving, storing and discharging through host organisms. Life both creates Force energy and channels it, and releases all its Force store to the greater Cosmic Force upon death. In life, the Cosmic Force provides via symbionts, and in receptive lifeforms the Force grows; the final crop is ultimately harvested by the Cosmic Force. A Godly composting and recycling system :)

I'm assuming the film title is only referring to a currently inactive living agent of the Force such as Luke, his apprentices or enemies, that they've resisted the Will of the Force for too long and by their inactivity, the polar opposite side of the Force has brought a critical level of inbalance?

Yep. I think to up the ante, there needs to be some great significance arising in the Force, something game-changing with profound galactic reverberations, or the film might just give us a run of the mill EU-type story. We've had six films describing probably the biggest social and spiritual upset in the Known Galaxy's history, and to tag on three more films that are just another go around the Maypole - blasters, starfighters, sass and Vader copycats - needs a heightened level of drama in spiritual and not just action-adventure terms especially since the Episodic saga is the flagship and captain of the spin-offs which don't need to be as 'standout' or mythic.

    Agreed +1000

I personally think this is absolutely correct thinking which is why I believe we will see Plagueis as a villain in that he achieved immortality. This level of supernatural ability surely causes a disruption or imbalance in the Force.

(Course I qualify my statement by admitting I still haven't seen S6 of TCW yet -- waiting for it to arrive. :p)
